The simplest format is simply @username++.

You'll get more value out of Karma bot environment when you accompany each karma request with the description.

@username ++ description requests positive karma for @username if it is allowed by Who can request karma option.

+1 karma requested for @username

If the requesting user is a moderator or Auto-approve all requests option is enabled there will be no such message.

To give more than 1 karma at once karma value should be specified within a command.

@username 5++ description requests karma 5 karma for @username.

+5 karma requested for @username

It is not possible to request karma for users who do not accept it. This is defined per user via Roles settings.

If a user does not accept karma Karma bot will give a warning.

Sorry, user @username does not accept karma.
